Heparin is a mucopolysaccharide that is widely present in mammalian tissues. It is mainly found in mast cells and has anticoagulant effects. It is widely used in surgery and treatment of cerebral thrombosis and myocardial infarction. Heparin sodium is the sodium salt of heparin, and as a natural anticoagulant substance, heparin sodium is valued by countries all over the world. It is also one of my country's main export drugs. With the deepening of research, people discovered that heparin sodium not only has anticoagulant, anti-thrombosis and blood lipid regulation effects, but also has anti-inflammatory, anti-allergic, anti-viral, and anti-cancer functions. At present, heparin sodium is mainly extracted from pig and sheep intestinal mucosa and bovine lungs. Studies have shown that heparin sodium has the highest content in pig intestinal mucosa. Crude heparin sodium is a traditional export product of my country and holds an important position in the world.

This article will introduce an extraction process of heparin sodium, which is simple to operate and has a high yield of heparin sodium, which is suitable for industrialized promotion and use, and specifically includes the following steps:

(1) Raw material processing: clean the fresh small intestine with clean water, scrape the small intestine mucosa after cleaning, and then put the small intestine mucosa in a blender until it becomes minced and set aside;

(2) Heating enzymatic hydrolysis: Put the minced intestinal mucosa obtained in step 1 into a heating tank, add water, mix and stir, and then add 8% trypsin and weak lye to adjust the pH value until the pH value of the solution is 8 -10, and then heat up. During the heating process, the pH should be kept between 8.5-9.5, heated to 30-40°C, kept at a constant temperature for 2-3h, and then continue to heat to 50-60°C, kept at the constant temperature for 10-20min, while Hot filtration to obtain filtrate;

(3) Cooling and adsorption: Cool the filtrate obtained in step 2, and remove the slick oil from the upper layer of the filtrate after cooling to 30-40°C, then add resin to stir and adsorb for 6-7h, and filter out the resin after the adsorption is complete;

(4) Elution: Put the collected resin into the eluent for washing, add 10% sodium chloride solution, keep the temperature at 50-55℃, stir for 3-4 hours, collect the eluate; Elute twice, combine the eluate collected twice for later use;

(5) Precipitation: Filter the eluate collected twice into the precipitation tank, add 80-85% alcohol with mass fraction and stir evenly, then adjust the pH value to keep at 7-8, and let stand for 10-12 hours for sealing and precipitation , Filter to get the precipitate for use;

(6) Dehydration and drying: Put the sediment in a drying oven to obtain heparin sodium.

As a preferred solution, the ratio of water to small intestinal mucosa in step (2) is 1:3.

As a preferred solution, the set temperature of the drying box is 50-60°C, and the drying time is 3-5h.
